SpecificityThis assay has high sensitivity and excellent specificity for detection of KLK10. No significant cross-reactivity or interference between KLK10 and analogues was observed.
-
Detection Range117.2-30,000pg/mL. The standard curve concentrations used for the CLIA's were 30,000pg/mL, 7,500pg/mL, 1,875pg/mL, 468.8pg/mL, 117.2pg/mL.
-
SensitivityThe minimum detectable dose of KLK10 is typically less than 41.3pg/mL. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean RLU value of 20 replicates of the zero standard subtracted by their two standard deviations.

Product Description specificalPrinciple of the assay: The microtiter plate provided in this kit has been pre-coated with a monoclonal antibody specific to KLK10. A competitive inhibition reaction is launched between biotin labeled KLK10 and unlabeled KLK10 (Standards or samples) with the pre-coated antibody specific to KLK10. After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of KLK10 in the sample. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is reverse proportional to the KLK10 level in the sample or standard.
