specificalIntroduction: Integrin-linked kinase (ILK) is a 59kDa protein originally identified while conducting a yeast-two hybrid screen with integrin beta1 as the bait protein (Hannigan et al., 1996). Since its discovery, ILK has been associated with multiple cellular functions including cell migration, cell proliferation, cell-adhesions, signal transduction. ILK1 is a member of the serine/threonine kinases, has been demonstrated to be associated with numerous biological and pathological processes. However, the clinical and functional significance of ILK1 expression has not been characterized previously in human astrocytoma. Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to ILK-1 has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with HRP-conjugated ILK-1 and incubated. A competitive inhibition reaction is launched between ILK-1 (Standards or samples) and HRP-conjugated ILK-1 with the pre-coated antibody specific for ILK-1. The more amount of ILK-1 in samples, the less antibody bound by HRP-conjugated ILK-1. Then the substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of ILK-1 in the sample. The color development is stopped and the intensity of the color is measured.
