    specifical    Introduction: The latent TGF-beta binding proteins (LTBP) are a family of carrier proteins. LTBP is a family of secreted multidomain proteins that were originally identified by their association with the latent form of transforming growth factors. They interact with a variety of extracellular matrix proteins and may play a role in the regulation of TGF beta bioavailability. LTBP-2 is an integral component of elastin-containing microfibrils. This protein is the largest member of the LTBP family possessing unique regions and with most similarity to the fibrillins. It has thus been suggested that it may have multiple functions: as a member of the TGF-beta latent complex, as a structural component of microfibrils, and a role in cell adhesion. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to LTBP2. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for LTBP2 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain LTBP2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of LTBP2 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
