Asprosin protein has been successfully developed

Baylor college of medicine researchers in the study of genetic disorder - type Neonatal premature aging disorder (NPS, Neonatal Progeroid Syndrome) of mechanism in the process, discovered a new type of glucose regulation protein hormone, named Asprosin.
In April 2016, the paper published in the Cell showed that Asprosin antibodies could regulate insulin levels in mice to normal, and long-term treatment could even restore insulin resistance in mice.
Asprosin's use in humans may provide a new treatment for diabetes.


In view of the application prospect of Asprosin, the protein department of our company has studied and produced the recombinant Asprosin.
We constructed the human FBN1 (2732-2871aa) gene into PET22b vector and expressed in e. coli.
The protein is composed of 140 objective amino acids and 6 of His. The molecular weight is 17KD.

We adopt the optimal expression condition and induce the expression of the recombinant His - asprosin e. coli bacteria.
The separation of escherichia coli from the Ni - NTA was combined with ni-nta and the method of affinity chromatography was used to purify His - asprosin protein.
The SDS - PAGE electrophoresis test was used to elute the samples. The experimental results showed that 50mM of imidazole was used to remove the target protein.

Asprosin of purified protein after further molecular sieves and to deal with the endotoxin, etc, and determined the purity (> 98%) and concentration (1 mg/ml), save in the PBS - glycerol buffer, can be used for other testing ELISA Kit and WB company.
